  • KLF5 (Krüppel-like factor 5) is a zinc finger transcription factor belonging to the Krüppel-like factor family, characterized by three highly conserved Cys2His2 zinc finger domains at its C-terminus that enable sequence-specific DNA binding to GC-rich elements in target gene promoters. This multifunctional protein plays pivotal roles in regulating cell proliferation, differentiation, apoptosis, and inflammation across various tissues, with its activity context-dependently acting as either an oncogene or tumor suppressor depending on the cellular environment and post-translational modifications. KLF5 is particularly significant in epithelial biology, where it maintains intestinal epithelial homeostasis, promotes vascular smooth muscle cell differentiation, and regulates adipogenesis. The protein undergoes extensive post-translational regulation, including acetylation, phosphorylation, ubiquitination, and SUMOylation, which modulate its stability, subcellular localization, and transcriptional activity. In cancer biology, KLF5 demonstrates complex dual functionality: it can promote tumorigenesis in prostate and breast cancers through activation of pro-survival genes, while exhibiting tumor-suppressive properties in colorectal and gastric cancers via regulation of cell cycle inhibitors like p21 and p27. Additionally, KLF5 serves as a critical mediator in multiple signaling pathways, including TGF-β, Wnt/β-catenin, and MAPK cascades, and has emerged as a potential therapeutic target and biomarker for various malignancies and inflammatory disorders, with ongoing research exploring its mechanistic roles in metabolic diseases and tissue regeneration.
